Key facts

Desloratadine is a non-sedating, long-acting H1 antihistamine used to relieve symptoms of allergic rhinitis and urticaria.

What it is used for

  • Symptoms of allergic rhinitis
  • Symptoms of urticaria
  • Film-coated tablets: adults and adolescents aged 12 years and older; oral solution: also children over the age of 1 year

Dosing

  • 5 mg film-coated tablets (adults and adolescents 12 years and over): one tablet once a day
  • 0.5 mg/ml oral solution: adults and adolescents 12 years and over 10 ml (5 mg) once a day
  • Oral solution: children 1 through 5 years 2.5 ml (1.25 mg), children 6 through 11 years 5 ml (2.5 mg) once a day
  • Intermittent allergic rhinitis: stop when symptoms resolve and restart if they reappear; persistent rhinitis: continued treatment during allergen exposure may be proposed
  • With or without food

Contraindications

  • Hypersensitivity to the active substance, any excipient or loratadine

Important warnings

  • Severe renal insufficiency: use with caution
  • Caution with a medical or family history of seizures, mainly in young children; consider stopping if a seizure occurs
  • Most rhinitis below 2 years of age is infectious; no data support treating it
  • Tablets contain lactose; oral solution contains sorbitol (not for hereditary fructose intolerance) and propylene glycol

Side effects

  • In trials reported in 3% of patients in excess of placebo; most frequent fatigue (1.2%), dry mouth (0.8%) and headache (0.6%)
  • Very rare: hallucinations, dizziness, somnolence, seizures, tachycardia, palpitations
  • Frequency not known: QT prolongation, increased appetite, abnormal behaviour

  • No clinically relevant interactions with erythromycin or ketoconazole in trials
  • Alcohol intolerance and intoxication reported post-marketing; caution with concomitant alcohol

Pregnancy and breastfeeding

  • A large amount of data indicates no malformative or foetal/neonatal toxicity; as a precaution, use during pregnancy is preferably avoided
  • Desloratadine has been found in breastfed infants; decide on stopping breast-feeding or therapy based on benefit
